Checking timming

In an effort to rule out an out of time motor ,I am going to attempt to check and adjust the timing with the motor in the car . I have bought a cam timing aliment tool . If I need to re-adjust timing , am I supposed to remove the tenisoner's and if so all or which one ? I read my vacuum gauge wrong and my motor is producing low vacuum the gauge says late timing . So I am assuming timing is off . I also bought an injector to put into cylinder 4 that has the miss .Injector should be here by weeks end . Thanks fr all your help .
