1) If you roll down a hill in neutral is the sound still there?

2) There could be a chip on the 1st drive gear.

3) Check to see if there is any damage to your driveshaft rubber boots - if there's a tear and water / debris got in, the bearing(s) could be on the way out.....

Of course, it could be something simple like a stone embedded in your tyre or brake disc, but if it only happens in 1st gear probably not......
