hard to say, but sounds like the regulator is starting to fail and the click is the wheel cocking a bit. Good news is the URO regulator isn't expensive, is easy to put in (with a few tricks) and works smooth with the ball bearing guide wheels. Taking off the door card you can check it the wheels seem wonky.
if you confirm that is what is wrong, search the forum a little for the tricks, and remember this:
The clamps with rubber lining that grab the window? Only loosen the bolts, don't remove them and make sure the rubber is separated on the new ones so a slot is there to put the window back into. Also pay attention to the limits set on the old one and duplicate this on the replacement part, you will get the adjustment pretty close this way, mine have always worked out perfect
